New Zealand blackcurrant extract modulates the heat shock response in men during exercise in hot ambient conditions.

To determine if 7d of New Zealand blackcurrant (NZBC) extract alters the heat shock, inflammatory and apoptotic response during prolonged exertional-heat stress. Ten men (Age: 29 ± 2years, Stature: 1.82 ± 0.02m, Mass: 80.3 ± 2.7kg, V̇O2max: 56 ± 2mL·kg-1·min-1) ingested two capsules of CurraNZ™ (NZBC extract: 210mg anthocyanins·day-1) or PLACEBO for 7d prior to 1h treadmill run (65% V̇O2max) in hot ambient conditions (34°C/40% RH). Blood samples were collected before (Pre), immediately after (Post), 1h after (1-Post), and 4h after (4-Post) exercise. Heat shock proteins (HSP90, HSP70, HSP32) were measured in plasma. HSP and protein markers of inflammatory capacity (TLR4, NF-κB) and apoptosis (BAX/BCL-2, Caspase 9) were measured in peripheral blood mononuclear cells (PBMC). eHSP32 was elevated at baseline in NZBC(+ 31%; p < 0.001). In PLACEBO HSP32 content in PBMC was elevated at 4-Post(+ 98%; p = 0.002), whereas in NZBC it fell at Post(-45%; p = 0.030) and 1-Post(-48%; p = 0.026). eHSP70 was increased at Post in PLACEBO(+ 55.6%, p = 0.001) and NZBC (+ 50.7%, p = 0.010). eHSP90 was increased at Post(+ 77.9%, p < 0.001) and 1-Post(+ 73.2%, p < 0.001) in PLACEBO, with similar increases being shown in NZBC (+ 49.0%, p = 0.006 and + 66.2%, p = 0.001; respectively). TLR4 and NF-κB were both elevated in NZBC at PRE(+ 54%, p = 0.003 and + 57%, p = 0.004; respectively). Main effects of study condition were also shown for BAX/BCL-2(p = 0.025) and Caspase 9 (p = 0.043); both were higher in NZBC. 7d of NZBC extract supplementation increased eHSP32 and PBMC HSP32 content. It also increased inflammatory and apoptotic markers in PBMC, suggesting that NZBCsupports the putative inflammatory response that accompanies exertional-heat stress.

The Effects of an Acute Dose of New Zealand Blackcurrant Extract on 5-km Running Performance.

This study investigated the effects of an acute dose (900mg) of New Zealand Blackcurrant (NZBC) extract on 5-km running performance, alongside associated physiological and metabolic responses. Sixteen trained male runners (age 26 ± 5years, stature 173.4 ± 7.3cm, body mass 73.7 ± 6.9 kg,maximal oxygen consumption [V˙O2max] 55.4 ± 6.1ml·kg-1·min-1) ingested either capsules containing NZBC extract (3 × 300mg CurraNZ, 315mg anthocyanins) or a matched placebo (3 × 300mg gluten-free flour) 2hr before exercise in a double-blind, randomized, crossover design. Performance time, physiological, and metabolic responses were assessed in a 5-km time trial, preceded by 10-min exercise at the lactate threshold on a treadmill. NZBC extract did not alter the physiological or metabolic responses to exercise at the lactate threshold (oxygen uptake, respiratory exchange ratio, minute ventilation, carbohydrate oxidation, fat oxidation, heart rate, blood lactate, or rating of perceived exertion, p > .05). The 5-km time trial was completed in a faster time in the NZBC extract condition compared with placebo (NZBC: 1,308.96 ± 122.36s, placebo: 1,346.33 ± 124.44, p = .001, d = -0.23, confidence interval range = [-0.46, 0.00s]). No differences in physiological or metabolic responses were apparent between conditions for the 5-km time trial (p > .05). Ingesting 900mg of NZBC extract as an acute dose improves performance in trained male runners without altering physiological or metabolic responses to exercise. Further research is needed to assess a wider range of possible mechanisms (e.g.,cardiovascular function, metabolite profiles) to advance insight into improved performance following supplementation.

Two weeks daily intake of anthocyanin-rich New Zealand blackcurrant extract enhances whole-body fat oxidation during supine rest in healthy males

Introduction: Increase in exercise-induced fat oxidation has been observed with a 2-week daily intake of New Zealand blackcurrant (NZBC) extract. We examined for supine rest the effects of a 2-week daily intake of NZBC extract on physiological and metabolic parameters in relation to body fat%.Methods: Healthy active men (n=16, age: 24.1±6.0 yr, body mass: 78.2±15.6 kg, BMI: 24.7±4.2 kg·m-2) volunteered. NZBC extract (210 mg of anthocyanins) was consumed with breakfast with a14-day washout. The final ingestion was 2 hours before testing. Physiological and metabolic responses were measured during supine rest (2x10 min), and observations with the lowest 10-minute ventilation were analyzed. Bioelectrical impedance analysis was used to measure body fat%.Results: During supine rest, a 14-day daily intake of NZBC extract had no effect on heart rate, minute ventilation, oxygen uptake, carbon dioxide production, and energy expenditure. However, a 14-day daily intake of NZBC extract decreased the respiratory exchange ratio (baseline: 0.840±0.045, 14-day: 0.820±0.058, P=0.03), increased fat oxidation (baseline: 0.079±0.03, 14-day: 0.088±0.043 g·min-1, P=0.05), and decreased carbohydrate oxidation (baseline: 0.178±0.067, 14-day: 0.143±0.071 g·min-1, P=0.03) during supine rest. Nine of the 16 participants had increased fat oxidation by over 10% during supine rest. For those nine participants, the increases were 26±16%. We did not observe a significant correlation between body fat% and absolute and percentual changes in fat oxidation.Conclusions: In a healthy male cohort, daily intake of New Zealand blackcurrant extract for two weeks was able to increase fat oxidation during supine rest. Daily intake of New Zealand blackcurrant extract may be an effective anthocyanin-rich supplement for managing body composition and body weight.Keywords: New Zealand blackcurrant; anthocyanins; metabolic equivalent; rest; fat oxidation; respiratory exchange ratio

Acute Effects of New Zealand Blackcurrant Extract on Cycling Time-Trial Are Performance Dependent in Endurance-Trained Cyclists: A Home-Based Study.

The intake of anthocyanin-rich New Zealand blackcurrant (NZBC) extract (300 mg per day) over a week enhanced 16.1 km cycling time trial (TT) performance in endurance-trained cyclists without acute performance effects. In the present study, the acute effects of an intake of 900 mg of NZBC extract 2 h before performing the 16.1 km cycling TT were examined. A total of 34 cyclists (26 males; 8 females) (age: 38 ± 7 years, V˙O2max: 57 ± 5 mL·kg-1·min-1) completed 4 16.1 km TTs (2 familiarization and 2 experimental trials) over 4 mornings on a home turbo-trainer connected with the online training simulator ZWIFT. There was no difference in time to complete the 16.1 km TT between conditions (placebo: 1422 ± 104 s; NZBC extract: 1414 ± 93 s, p = 0.07). However, when participants were split between faster (<1400 s; 1 female; 16 males) and slower (>1400 s; 7 females; 10 males) cyclists based on average familiarization TTs, a difference in TT performance was observed only in the slower group (placebo: 1499 ± 91 s; NZBC extract: 1479 ± 83 s, p = 0.02). At 12 km (quartile analysis), power output (p = 0.04) and speed (p = 0.04) were higher compared to the placebo with no effects on heart rate and cadence. The acute effects of 900 mg of NZBC extract on a 16.1 km cycling time-trial may depend on the performance ability of male endurance-trained cyclists. More work is needed to address whether there is a sex-specific time-trial effect of NZBC extract independent of performance ability.

Acute Consumption of New Zealand Blackcurrant Extract Has No Effect on Cycling Performance in Normobaric Hypoxia with Trained Cyclists

Introduction: New Zealand Blackcurrant Extract (NZBC) is a popular ergogenic aid used to improve endurance performance. The aim of this research was to determine the effects of a single bolus of NZBC on 10-km time trial (TT) cycling performance in normobaric hypoxia. Methods: A double-blind, crossover design study was conducted with trained cyclists. The effects of acute NZBC (900 mg) were compared with a placebo in normobaric hypoxia (NH) (FiO2 = 15.5%). Testing comprised of three laboratory-based visits for (1) familiarisation (and screening of TT performance before entry into study), (2) placebo and (3) NZBC, whereby a 10-km cycling TT was completed one hour after consumption. After completion of the TT blood lactate was assessed at four time-points in the 10 minutes following. Throughout the TT, power output (PO), rating of perceived exertion (RPE) and heart rate (HR) were recorded. Results: NZBC had no effect on TT cycling performance in NH compared to a placebo (1078.4 s [1009.4, 1147.4] and 1071.0 s [1006.4, 1137.5] respectively, p=0.31; d=-0.31). Additionally, no difference was observed for mean power output (p=0.20; d=0.39), HR (p=0.76; d=0.09) or at 1-km intervals for performance time (p=0.80), PO (p=0.77) or RPE (p=0.41). Post exercise blood lactate recovery did not differ between placebo and NZBC (p=0.42). Conclusion: Acute intake of NZBC has no effect on cycling performance or blood lactate recovery in simulated altitude.

Running-Induced Metabolic and Physiological Responses Using New Zealand Blackcurrant Extract in a Male Ultra-Endurance Runner: A Case Study.

Physical training for ultra-endurance running provides physiological adaptations for exercise-induced substrate oxidation. We examined the effects of New Zealand blackcurrant (NZBC) extract on running-induced metabolic and physiological responses in a male amateur ultra-endurance runner (age: 40 years, body mass: 65.9 kg, BMI: 23.1 kg·m−2, body fat: 14.7%, V˙O2max: 55.3 mL·kg−1·min−1, resting heart rate: 45 beats·min−1, running history: 6 years, marathons: 20, ultra-marathons: 28, weekly training distance: ~80 km, weekly running time: ~9 h). Indirect calorimetry was used and heart rate recorded at 15 min intervals during 120 min of treadmill running (speed: 10.5 km·h−1, 58% V˙O2max) in an environmental chamber (temperature: ~26 °C, relative humidity: ~70%) at baseline and following 7 days intake of NZBC extract (210 mg of anthocyanins·day−1) with constant monitoring of core temperature. The male runner had unlimited access to water and consumed a 100-kcal energy gel at 40- and 80 min during the 120 min run. There were no differences (mean of 8, 15 min measurements) for minute ventilation, oxygen uptake, carbon dioxide production and core temperature. With NZBC extract, the respiratory exchange ratio was 0.02 units lower, carbohydrate oxidation was 11% lower and fat oxidation was 23% higher (control: 0.39 ± 0.08, NZBC extract: 0.48 ± 0.12 g·min−1, p < 0.01). Intake of the energy gel did not abolish the enhanced fat oxidation by NZBC extract. Seven days’ intake of New Zealand blackcurrant extract altered exercise-induced substrate oxidation in a male amateur ultra-endurance runner covering a half-marathon distance in 2 h. More studies are required to address whether intake of New Zealand blackcurrant extract provides a nutritional ergogenic effect for ultra-endurance athletes to enhance exercise performance.

Effects of New Zealand Blackcurrant Extract on Sequential Performance Testing in Male Rugby Union Players.

Previous studies on performance effects by New Zealand blackcurrant (NZBC) extract used mainly a single exercise task. We examined the effects of NZBC extract in a battery of rugby union–specific tests including speed, agility and strength testing. University male rugby union players (n = 13, age: 21 ± 2 years, height: 182 ± 6 cm, body mass: 87 ± 13 kg) completed two full familiarisations and two experimental visits in an indoor facility. The study had a double blind, placebo-controlled, randomised, crossover design. For the experimental visits, participants consumed NZBC extract (210 mg/day of anthocyanins for 7 days) or placebo with a 7-day wash-out. Testing order was the running-based anaerobic sprint test, the Illinois agility test, seated medicine ball (3 kg) throw, and handgrip strength. With NZBC extract, there may have been an effect for average sprint time to be faster by 1.7% (placebo: 5.947 ± 0.538 s, NZBC extract: 5.846 ± 0.571 s, d = −0.18 (trivial), p = 0.06). However, with NZBC extract there may have been reduced slowing of sprint 2 (d = −0.59 (moderate), p = 0.06) and reduced slowing for sprint 6 (d = −0.56 (moderate), p = 0.03). In the Illinois agility test, there may have also been an effect for the mean time to be faster by 1.6% (placebo: 18.46 ± 1.44 s, NZBC extract: 18.15 ± 1.22 s, d = −0.24 (small), p = 0.07). The correlation between the %change in average sprint time and %change in mean agility time was not significant (Pearson R2 = 0.0698, p = 0.383). There were no differences for the seated medicine ball throw distance (p = 0.106) and handgrip strength (p = 0.709). Intake of NZBC extract in rugby union players seems to improve tasks that require maximal speed and agility but not muscle strength. NZBC blackcurrant extract may be able to enhance exercise performance in team sports that require repeated movements with high intensity and horizontal change of body position without affecting muscle strength.

Effect of New Zealand Blackcurrant Extract on Force Steadiness of the Quadriceps Femoris Muscle during Sustained Submaximal Isometric Contraction.

Intake of anthocyanin-rich New Zealand blackcurrant (NZBC) can alter physiological responses that enhance exercise performance. In two studies, we examined the effects of NZBC extract on force steadiness during a sustained submaximal isometric contraction of the quadriceps femoris muscle. With repeated measures designs, male participants in study one (n = 13) and study two (n = 19) performed a 120 s submaximal (30%) isometric contraction of the quadriceps femoris muscle following a 7-day intake of NZBC extract and placebo (study one) and following 0 (control), 1-, 4- and 7-day intake of NZBC extract (study two). Participants for both studies were different. In study one, NZBC extract enhanced isometric force steadiness during the 120 s contraction (placebo: 6.58 ± 2.24%, NZBC extract: 6.05 ± 2.24%, p = 0.003), with differences in the third (60–89 s) and fourth quartile (90–120 s) of the contraction. In study two, isometric force steadiness was not changed following 1 and 4 days but was enhanced following 7-day intake of NZBC extract in comparison to control. In study two, the enhanced isometric force steadiness following 7-day intake did occur in the second (30–59 s), third (60–89 s) and fourth (90–120 s) quartiles. Daily supplementation of anthocyanin-rich NZBC extract can enhance force steadiness of the quadriceps femoris muscle during a sustained submaximal isometric contraction. Our observations may have implications for human tasks that require postural stability.

Enhanced Walking-Induced Fat Oxidation by New Zealand Blackcurrant Extract Is Body Composition-Dependent in Recreationally Active Adult Females.

New Zealand blackcurrant (NZBC) extract enhanced cycling-induced fat oxidation in female endurance athletes. We examined in recreationally active females the effects of NZBC extract on physiological and metabolic responses by moderate-intensity walking and the relationship of fat oxidation changes with focus on body composition parameters. Twelve females (age: 21 ± 2 y, BMI: 23.6 ± 3.1 kg·m−2) volunteered. Bioelectrical bioimpedance analysis was used for body composition measurements. Resting metabolic equivalent (1-MET) was 3.31 ± 0.66 mL·kg−1·min−1. Participants completed an incremental walking test with oxygen uptake measurements to individualize the treadmill walking speed at 5-MET. In a randomized, double-blind, cross-over design, the 30 min morning walks were in the same phase of each participant’s menstrual cycle. No changes by NZBC extract were observed for walking-induced heart rate, minute ventilation, oxygen uptake, and carbon dioxide production. NZBC extract enhanced fat oxidation (10 responders, range: 10–66%). There was a significant correlation for changes in fat oxidation with body mass index; body fat% in legs, arms, and trunk; and a trend with fat oxidation at rest but not with body mass and habitual anthocyanin intake. The NZBC extract responsiveness of walking-induced fat oxidation is body composition-dependent and higher in young-adult females with higher body fat% in legs, arms, and trunk.

Intake Duration of Anthocyanin-Rich New Zealand Blackcurrant Extract Affects Cardiovascular Responses during Moderate-Intensity Walking But Not at Rest

We examined effects of intake duration of New Zealand blackcurrant (NZBC) extract on cardiovascular responses during supine rest and moderate-intensity walking. Recreationally active men (n = 15, age: 24 ± 6 yr, BMI: 24.7 ± 4.3 kg·m−2) volunteered in a randomized, cross-over design. One metabolic equivalent (1-MET) was measured (3.97 ± 0.66 mL·kg−1·min−1) and an incremental walking test was performed to individualize speed at 4 (n = 3) or 5 (n = 12) METs for the 30-min walk (5.7 ± 0.7 km·hr−1). NZBC extract (210 mg of anthocyanins) was taken with breakfast for 7 and 14 days, with a 14-days washout. The final dose was ingested 2-hr before recording of the cardiovascular responses (Portapres Model-2). At rest, %changes at 7- and 14-days intake were observed for stroke volume (+6.8% (trend), p = 0.065; +8.5%, p = 0.012), cardiac output (+10.1%, p = 0.007; +8.5%, p = 0.013), total peripheral resistance (−12.0%, p = 0.004; −13.1%, p = 0.011), diastolic (−5.7%, p = 0.045; −9.7%, p = 0.015) and mean arterial pressure (−4.4%, p = 0.040; −7.2%, p = 0.029), but without intake duration effect. During walking, %changes at 7- and 14-days intake were observed for stroke volume (+7.7% (trend), p = 0.063; +9.9%, p = 0.006), cardiac output (+8.7%, p = 0.037; +10.1%, p = 0.007), diastolic blood pressure (−6.2%, p = 0.042; −10.6%, p = 0.001), and total peripheral resistance (−9.6%, p = 0.042; −13.5%, p = 0.005) but without intake duration effect. During walking, %changes at 14-days were observed only for mean arterial pressure (−6.4%, p = 0.018) and arterio-venous oxygen difference (−7.9%, p = 0.019). NZBC extract affects cardiovascular responses at rest and during moderate-intensity exercise with 7- and 14-day intake. Only during moderate-intensity exercise, a longer intake of NZBC extract was required for an effect on some cardiovascular responses.

Consumption of New Zealand Blackcurrant Extract Improves Recovery from Exercise-Induced Muscle Damage in Non-Resistance Trained Men and Women: A Double-Blind Randomised Trial.

Background: Blackcurrant is rich in anthocyanins that may protect against exercise-induced muscle damage (EIMD) and facilitate a faster recovery of muscle function. We examined the effects of New Zealand blackcurrant (NZBC) extract on indices of muscle damage and recovery following a bout of strenuous isokinetic resistance exercise. Methods: Using a double-blind, randomised, placebo controlled, parallel design, twenty-seven healthy participants received either a 3 g·day−1 NZBC extract (n = 14) or the placebo (PLA) (n = 13) for 8 days prior to and 4 days following 60 strenuous concentric and eccentric contractions of the biceps brachii muscle on an isokinetic dynamometer. Muscle soreness (using a visual analogue scale), maximal voluntary contraction (MVC), range of motion (ROM) and blood creatine kinase (CK) were assessed before (0 h) and after (24, 48, 72 and 96 h) exercise. Results: Consumption of NZBC extract resulted in faster recovery of baseline MVC (p = 0.04), attenuated muscle soreness at 24 h (NZBC: 21 ± 10 mm vs. PLA: 40 ± 23 mm, p = 0.02) and 48 h (NZBC: 22 ± 17 vs. PLA: 44 ± 26 mm, p = 0.03) and serum CK concentration at 96 h (NZBC: 635 ± 921 UL vs. PLA: 4021 ± 4319 UL, p = 0.04) following EIMD. Conclusions: Consumption of NZBC extract prior to and following a bout of eccentric exercise attenuates muscle damage and improves functional recovery. These findings are of practical importance in recreationally active and potentially athletic populations, who may benefit from accelerated recovery following EIMD.

Effect of Intake Duration of Anthocyanin-Rich New Zealand Blackcurrant Extract on Cardiovascular Responses and Femoral Artery Diameter during Sustained Submaximal Isometric Contraction

Seven-day intake of anthocyanins from New Zealand blackcurrant (NZBC) extract increased cardiac output and femoral artery diameter during a sustained submaximal isometric contraction. It is not known if there are intake duration effects by NZBC extract on the isometric contraction-induced cardiovascular responses. In a repeated measures design, male participants (n = 19, age: 26 ± 4 years) performed a 120-second submaximal (30%) isometric contraction of the knee extensors at baseline and following 1, 4 and 7-days intake of 600 mg·day−1 NZBC extract. During the 120-second submaximal isometric contraction, femoral artery diameter and cardiovascular responses were measured with ultrasound and beat-to-beat hemodynamic monitoring. Femoral artery was larger following 4-days (mean difference = 0.046 cm, 95% CI [0.012, 0.080], p = 0.005) and 7-days (mean difference = 0.078 cm, 95% CI [0.034, 0.123], p < 0.001) in comparison to baseline with no increase with 1-day intake. Systolic and diastolic blood pressure, heart rate and total peripheral resistance were not changed by NZBC extract at 1, 4 and 7-days intake. However, mean arterial pressure, stroke volume, cardiac output and total peripheral resistance were changed at time points during the isometric contraction following 7-days intake in comparison to 1-day intake of NZBC extract (p < 0.05). Alterations in femoral artery diameter and some cardiovascular responses during a submaximal sustained isometric contraction of the knee extensors are affected by the intake duration of New Zealand blackcurrant extract, with no effects by 1-day intake. Our observations suggest that the bioavailability of blackcurrant anthocyanins and anthocyanin-derived metabolites is required for days to alter the mechanisms for isometric-contraction induced cardiovascular responses.

Plasma uptake of selected phenolic acids following New Zealand blackcurrant extract supplementation in humans

New Zealand blackcurrant (NZBC) extract is a rich source of anthocyanins and in order to exert physiological effects, the anthocyanin-derived metabolites need to be bioavailable in vivo. We examined the plasma uptake of selected phenolic acids following NZBC extract supplementation alongside maintaining a habitual diet (i.e. not restricting habitual polyphenol intake). Twenty healthy volunteers (nine females, age: 28 ± 7 years, height 1.73 ± 0.09 m, body mass 73 ± 11 kg) consumed a 300 mg NZBC extract capsule (CurraNZ®; anthocyanin content 105 mg) following an overnight fast. Venous blood samples were taken pre and 1, 1.5, 2, 3, 4, 5, and 6 h post-ingestion of the capsule. Reversed-phase high-performance liquid chromatography (HPLC) was used for analysis of two dihydroxybenzoic acids [i.e. vanillic acid (VA) and protocatechuic acid (PCA)] and one trihydroxybenzoic acid [i.e. gallic acid (GA)] in plasma following NZBC extract supplementation. Habitual anthocyanin intake was 168 (95%CI:68–404) mg⋅day−1 and no associations were observed between this and VA, PCA, and GA plasma uptake by the NZBC extract intake. Plasma time-concentration curves revealed that GA, and PCA were most abundant at 4, and 1.5 h post-ingestion, representing a 261% and 320% increase above baseline, respectively, with VA remaining unchanged. This is the first study to demonstrate that an NZBC extract supplement increases the plasma uptake of phenolic acids GA, and PCA even when a habitual diet is followed in the days preceding the experimental trial, although inter-individual variability is apparent.

New Zealand blackcurrant extract enhances muscle oxygenation during repeated intermittent forearm muscle contractions in advanced and elite rock climbers

Anthocyanin-rich New Zealand blackcurrant (NZBC) may improve forearm muscle oxygenation and enhance performance in high-level rock climbers. As such, using a double-blind, randomised, cross-over design study, twelve participants performed an oxidative capacity assessment, and two successive exhaustive exercise trials (submaximal forearm muscle contractions at 60% of their maximal volitional contraction). Each visit was conducted following 7-days intake of 600 mg·day−1 NZBC extract or placebo. Oxidative capacity was estimated by calculating the oxygen half time recovery using near infrared spectroscopy. Time to exhaustion (s), impulse (kg·s), and minimum tissue saturation index (min-TSI %) were assessed during both the exercise trials. Muscle oxidative capacity was greater with NZBC (mean difference [MD] = 5.3 s, 95% confidence intervals [95% CI] = 0.4–10.2 s; p = 0.036; Cohen’s d = 0.94). During the exercise trials, there was an interaction for min-TSI % (time x condition, p = 0.046;  = 0.372), which indicated a greater level of oxygen extraction during trial two with NZBC extract (MD = 9%, 95% CI = 2-15%) compared to the placebo (MD = 2%, 95% CI = 1–7%). There was a decrease in time to exhaustion (p <0.001,  = 0.693) and impulse (p = 0.001,  = 0.672) in exercise trial two, with no effect of NZBC extract. In high-level rock climbers 7-days NZBC extract improves forearm muscle oxygenation with no effect on isolated forearm muscle performance.

No Effects of Different Doses of New Zealand Blackcurrant Extract on Cardiovascular Responses During Rest and Submaximal Exercise Across a Week in Trained Male Cyclists.

Supplementation with anthocyanin-rich blackcurrant increases blood flow, cardiac output, and stroke volume at rest. It is not known whether cardiovascular responses can be replicated over longer timeframes in fed trained cyclists. In a randomized, double-blind, crossover design, 13 male trained cyclists (age 39 ± 10 years, V˙O2max 55.3 ± 6.7ml·kg-1·min-1) consumed two doses of New Zealand blackcurrant (NZBC) extract (300 and 600mg/day for 1 week). Cardiovascular parameters were measured during rest and submaximal cycling (65% V˙O2max) on day 1 (D1), D4, and D7. Data were analyzed with an RM ANOVA using dose (placebo vs. 300 vs. 600mg/day) by time point (D1, D4, and D7). Outcomes from placebo were averaged to determine the coefficient of variation within our experimental model, and 95% confidence interval (CI) was examined for differences between placebo and NZBC. There were no differences in cardiovascular responses at rest between conditions and between days. During submaximal exercise, no positive changes were observed on D1 and D4 after consuming NZBC extract. On D7, intake of 600mg increased stroke volume (3.08ml, 95% CI [-2.08, 8.26]; d = 0.16, p = .21), cardiac output (0.39L/min, 95% CI [-1.39, .60]; d = 0.14, p = .40) (both +2.5%), and lowered total peripheral resistance by 6.5% (-0.46mmHg·min/ml, 95% CI [-1.80, .89]; d = 0.18, p = .46). However, these changes were trivial and fell within the coefficient of variation of our study design. Therefore, we can conclude that NZBC extract was not effective in enhancing cardiovascular function during rest and submaximal exercise in endurance-trained fed cyclists.

Daily and Not Every-Other-Day Intake of Anthocyanin-Rich New Zealand Blackcurrant Extract Alters Substrate Oxidation during Moderate-Intensity Walking in Adult Males

Daily intake of anthocyanin-rich New Zealand blackcurrant (NZBC) extract can enhance exercise-induced fat oxidation. It is not known whether habitual dietary anthocyanin intake and body composition affects blackcurrant-induced fat oxidation or even if daily intake is required. We examined effects of daily and every-other-day intake of NZBC extract on metabolic and physiological responses during moderate-intensity walking. Sixteen physically active males (age: 24 ± 6 yr, body mass: 78 ± 16 kg, BMI: 24.7 ± 4.2 kg·m −2, body fat: 15.2 ± 5.0%) volunteered. A randomized, cross-over design with a control condition was used and habitual dietary anthocyanin intake quantified. For intake conditions, participants consumed two capsules of NZBC extract (i.e. 210 mg of anthocyanins, CurraNZ™) with breakfast daily or every-other-day for 14 days (14-D and 14-EOD) with 14-days washout. Final two capsules were taken 2-hr before the walk (speed: 5.7 ± 0.7 km·hr−1). There was a trend for lower respiratory exchange ratio and carbohydrate oxidation with changes only for 14-D. Fat oxidation was increased only for 14-D (p  < 0.05) with 50% of the participants more than a 10% change. In 14-D, there was a positive correlation for BMI and body fat % with the absolute change in fat oxidation but not with habitual dietary anthocyanin intake. Daily intake of NZBC extract is required to enhance exercise-induced fat oxidation. Enhanced exercise-induced fat oxidation by daily intake of NZBC extract is related to body composition but not to habitual dietary anthocyanin intake in physically active males. Daily anthocyanin intake seems to be required to allow the gradual build-up and maintenance of anthocyanin-derived metabolites that are required to alter mechanisms for exercise-induced substrate oxidation.

Effect of New Zealand Blackcurrant Extract on Isometric Contraction-Induced Fatigue and Recovery: Potential Muscle-Fiber Specific Effects.

New Zealand blackcurrant (NZBC) extract has shown performance-enhancing effects during cycling, running and sport climbing. We examined effects of NZBC extract on (1) voluntary and twitch force of the quadriceps femoris muscles during repeated isometric contraction-induced fatigue, (2) twitch force during recovery and (3) muscle fiber-specific effects. Familiarized recreationally active males (n = 12, age: 24 ± 5 yrs; height: 180 ± 5 cm; body mass: 89 ± 11 kg) performed sixteen, 5-s voluntary maximal isometric contractions (iMVC) separated by 3-s rest. Twitch force was recorded before, during the 3-s rests and 5-min recovery. Supplementation consisted of 7-days intake of NZBC extract (600 mg∙day−1 containing 210 mg anthocyanin) in a double-blind, randomized, placebo-controlled crossover design with a 14-days washout. NZBC extract allowed for greater force in the first quartile of the iMVCs. Twitch force at baseline was 12% higher with NZBC extract (p = 0.05). However, there was no effect of NZBC for twitch force during the 16-iMVCs and recovery. Based on the maximum post-activation potentiation during the placebo 16-iMVCs, four subjects were classified of having a predominant type I or II muscle fiber typology. In type II, NZBC extract provided a trend for increased MVC force (~14%) in the first quartile and for type I in the fourth quartile (~10%). In type I, NZBC extract seemed to have higher twitch forces during the fatiguing exercise protocol and recovery, indicating increased fatigue resistance. New Zealand blackcurrant extract affects force during repeated maximal isometric contractions. Future work on mechanisms by NZBC extract for muscle fiber-specific fatigue-induced force responses is warranted.

No Effects of New Zealand Blackcurrant Extract on Physiological and Performance Responses in Trained Male Cyclists Undertaking Repeated Testing across a Week Period.

Anthocyanin supplements are receiving attention due to purported benefits to physiological, metabolic, and exercise responses in trained individuals. However, the efficacy of anthocyanin intake over multiple testing days is not known. We compared a placebo and two doses of anthocyanin-rich New Zealand blackcurrant (NZBC) extract (300 and 600 mg·day−1) on plasma lactate, substrate oxidation, and 16.1 km time trial (TT) performance on three occasions over 7-days in a fed state (day 1 (D1), D4, and D7). Thirteen male cyclists participated in a randomized, crossover, placebo-controlled double-blind design. There was no difference in plasma lactate and substrate oxidation between conditions and between days. A time difference was observed between D1 (1701 ± 163 s) and D4 (1682 ± 162 s) for 600 mg (p = 0.05), with an increment in average speed (D1 = 34.3 ± 3.4 vs. D4 = 34.8 ± 3.4 km·h−1, p = 0.04). However, there was no difference between the other days and between conditions. Overall, one week of intake of NZBC extract did not affect physiological and metabolic responses. Intake of 600 mg of NZBC extract showed inconsistent benefits in improving 16.1 km time trial performance over a week period in trained fed cyclists.

Short-term, but not acute, intake of New Zealand blackcurrant extract improves insulin sensitivity and free-living postprandial glucose excursions in individuals with overweight or obesity

Impaired postprandial glucose handling and low-grade systemic inflammation are risk factors for developing insulin resistance in individuals with overweight or obesity. Acute ingestion of anthocyanins improves postprandial glucose responses to a single carbohydrate-rich meal under strictly controlled conditions.PurposeExamine whether acute and short-term supplementation with anthocyanin-rich New Zealand blackcurrant (NZBC) extract can improve postprandial glucose responses to mixed-macronutrient meals.MethodsTwenty-five overweight (BMI > 25 kg m2) sedentary individuals participated in one of the following double-blinded, randomised controlled trials: (1) ingestion of 600 mg NZBC extract or placebo prior to consumption of a high-carbohydrate, high-fat liquid meal (n = 12); (2) 8-days supplementation with NZBC extract (600 mg day−1) or placebo, with insulin sensitivity and markers of inflammation assessed on day-7, and free-living postprandial glucose (continuous glucose monitoring) assessed on day-8 (n = 13).ResultsA single dose of NZBC extract had no effect on 3 h postprandial glucose, insulin or triglyceride responses. However, in response to short-term NZBC extract supplementation insulin sensitivity was improved (+ 22%; P = 0.011), circulating C-reactive protein concentrations decreased (P = 0.008), and free-living postprandial glucose responses to both breakfast and lunch meals were reduced (− 9% and − 8%, respectively; P < 0.05), compared to placebo.ConclusionThese novel results indicate that repeated intake, rather than a single dose of NZBC extract, is required to induce beneficial effects on insulin sensitivity and postprandial glucose handling in individuals with overweight or obesity. Continuous glucose monitoring enabled an effect of NZBC extract to be observed under free-living conditions and highlights the potential of anthocyanin-rich supplements as a viable strategy to reduce insulin resistance.

Intake Duration of Anthocyanin-Rich New Zealand Blackcurrant Extract Affects Metabolic Responses during Moderate Intensity Walking Exercise in Adult Males

We examined effects of intake duration of anthocyanin-rich New Zealand blackcurrant (NZBC) extract on physiological and metabolic responses during moderate intensity walking. Healthy men (n = 16, age: 24 ± 6 years, body mass: 78 ± 16 kg, BMI: 24.7 ± 4.1 kg·m −2, body fat: 15 ± 5%) volunteered. One metabolic equivalent (1-MET: 3.95 ± 0.64 ml·kg−1·min−1) was measured during supine rest. Responses during the 30-min walk (n = 3: 4-MET; n = 13: 5-MET) (speed: 5.7 ± 0.7 km·hr−1) were measured at 7-10, 17-20 and 27-30 min and averaged over the time periods. For intake conditions (7-days and 14-days), two capsules of NZBC extract (600 mg containing 210 mg of anthocyanins) were taken with breakfast (14-day washout). The final two capsules were ingested 2-hr before the morning walk. Intake duration of NZBC extract had no effect on heart rate, minute ventilation, oxygen uptake, and carbon dioxide production. Fat oxidation was enhanced with 7- and 14-day intake by 11 ± 19% and 17 ± 26% (baseline: 0.36 ± 0.12, 7-day: 0.39 ± 0.13, 14-day: 0.41 ± 0.13 g·min−1, p = 0.007). Only 14-day intake lowered RER (baseline: 0.852 ± 0.046, 7-day: 0.843 ± 0.045, 14-day: 0.837 ± 0.037, p = 0.019) and carbohydrate oxidation (baseline: 0.95 ± 0.40, 7-day: 0.91 ± 0.40, 14-day: 0.86 ± 0.33 g·min−1, p = 0.032). Rating of perceived exertion was lower with 7-day and 14-day intake (baseline: 11.0 ± 2.3, 7-day: 10.5 ± 1.8, 14-day: 10.3 ± 2.1, p = 0.002). Longer intake duration (i.e. 14 days) of New Zealand blackcurrant extract seems to enhance fat oxidation more during a 30-min moderate intensity walk than 7 days intake. The intake duration of anthocyanin-rich New Zealand blackcurrant extract may be due to an enhanced bioavailability of anthocyanin-derived metabolites that alter the mechanisms for substrate oxidation during moderate intensity exercise.
